CpG-ODN刺激HBV免疫清除期及耐受期患者PBMCs分泌IFNɑ的研究

摘　　要：目的研究免疫清除期、免疫耐受期HBV感染者外周血单个核细胞(PBMCs)对寡核苷酸CpG-ODN刺激的差异。方法 HBV感染免疫清除期与免疫耐受期患者32例,常规分离PBMCs,体外用CpG-ODN刺激,用酶联免疫斑点(ELISPOT)方法检测PBMCs分泌的IFNɑ。结果 100.0%的免疫清除期HBV感染者PBMCs受CpG-ODN刺激、IFNɑ分泌实验阳性,而免疫耐受期的HBV感染者PBMCs受CpG-ODN刺激,只有40.0%IFNɑ分泌实验阳性,二者之间差异有统计学意义(P<0.01)。结论免疫清除期、免疫耐受期HBV感染者PBMCs对CpG-ODN刺激的免疫反应不同。OBJECTIVE To study the different response of peripheral blood mononucla cells（PBMCs）stimulated by CpG-ODN between the patients infected with HBV in clearance period and those in tolerance period.METHODS Forty-two patients chronically infected with HBV,22 patients in clearance period,and 20 patients in tolerance period were included.The PBMCs from the patients got by normal method were stimulated with CpG-ODN separately in vitro.The production of IFNɑwere measured by the method of enzyme-linked immunospot（ELISPOT）.RESULTS In compared with the production of IFNɑ stimulated with the culture media 1640,those stimulated with CpG-ODN were significant higher for the 100.0% of patients in clearance period and 40.0% of the patients in tolerance period,there was significant difference between them.CONCLUSIONS The response of the production of IFNɑ stimulated with CpG-ODN for the patients in clearance period is significant different from those in tolerance period.
