摘 要:目的观察眼镜蛇毒因子(CVF)对慢性坐骨神经损伤(CCI)后大鼠脊髓背角神经元的保护效应。方法 36只SD大鼠随机均分成三组,CCI+生理盐水(CS组)、CCI+CVF 50μg/kg(CC组)和尾静脉注射生理盐水(SS组)。于术前和术后1、3、5、7 d测定大鼠的热痛阈值和机械痛阈值,并于术后7 d处死大鼠取脊髓组织,在电镜下观察脊髓背角神经元的内部结构变化,并测定脊髓匀浆中超氧化物歧化酶(SOD)和丙二醛(MDA)吸光度。结果与SS组相比,CS组大鼠热痛阈值和机械痛阈值降低,脊髓匀浆中SOD活力明显降低,而MDA显著升高(P<0.05);与CS组相比,CC组大鼠热痛阈值和机械痛阈值升高,脊髓匀浆中SOD活力明显升高,而MDA却迅速下降(P<0.01)。给予CVF后,CCI大鼠脊髓背角神经元线粒体的肿胀程度减轻。结论 CVF能减轻CCI后脊髓背角神经元的损伤程度,可能与其减少自由基和脂质过氧化的损伤有关。Objective To observe the protective effect of cobra venom factor on neurons in spinal dorsal horn in rats with neuropathic pain(NPP). Methods SD rats were divided randomly into three groups of sham operation(SS), CCI+saline(CS) and CCI+CVF50 g/k(CC). Sciatic nerve ligature was performed classically in groups of CS and CC. Normal saline and CVF were injected daily through caudal vein. Pain threshold of thermal and mechanical stimulation was recorded before and on the 1^st, 3^rd, 5^th and 7^th clay after operation The internal structure of neuron in dorsal horn was observed on the 7th day after operation by eleetronmicroseopy. While the levels of superoxide dismutase(SOD) and malondialdehyde (MDA) in the spinal were determined with colorimetric method. Results Compared with group SS, thermal and mechanical pain thresholds were significantly decreased, the activity of SOD was reduced obviously, and the level of MDA was raised obviously in groupCS. Compared with rats in group CS, the pain threshold was obversly higher, the activity of SOD was recovered in a large degree, and the level of MDA was decreased in spinal cord in group CC. Intact structures and no swelling of such organdies as mitochondria in spinal dorsal horn neurons were seen in group SS, but mitochondrial swelling, crista disruption and cytomembrane breakdown of spinal dorsal horn neurons could be seen in group CS, which were significantly less in group CC. Conclusion CVF can effectively protect neurons in spinal dorsal horn in rats with NPP,which may be related to its effect of reducing the injury of free radical and lipid peroxidation.
