摘　　要：目的观察戊二醛对静脉移植物内膜增生的影响。方法60只新西兰大白兔随机分为实验组和对照组，实验组静脉移植物用1％戊二醛溶液浸泡10min，对照组用生理盐水浸泡10min。分别于移植后4、12周时观察内膜增生情况；对平滑肌细胞肌动蛋白α（α-actin）、增殖细胞核抗原（PCNA）行免疫组织化学染色；行原位末端标记法（TUNEL）染色观察细胞凋亡。结果术后4周和12周，实验组平滑肌细胞明显少于对照组，内膜厚度（0．016±0．004）、（0．031±0．008）min小于对照组（0．025±0．005）、（0．093±0．024）mitt（P〈0．05），但两组细胞增殖率差异无统计学意义（P〉0．05）。细胞凋亡率实验组（3．3±0．7）％、（6．7±1．5）％高于对照组（1．2±0．4）％、（1．8±0．5）％（P〈0．05）。结论戊二醛处理静脉移植物可显著减少移植物平滑肌细胞数目并抑制内膜增生。Objective To investigate the effect of treatment of jugular vein graft with glutaraldehyde on intimal hyperplasia in rabbits. Methods Sixty New Zealand white rabbits were randomly divided into experimental group and the control group. Before the anastomosis, the vein in the experimental group was soaked in 1% glutaraldehyde solution for 10 min, and that in control group in normal saline for 10 min. The vein graft was harvested after 4 and 12 weeks. HE staining, smooth muscle cell α-actin and proliferating cell nuclear antigen （PCNA） immunohistochemical staining were performed. Apoptosis was detected by TUNEL. Results Four and 12 weeks after grafting, the number of smooth muscle cells in the experimental group was significantly less than that in the control group, and intima thickness [ （0. 016 ± 0. 004）, （0. 031 ± 0. 008 ）mm ] in the experimental group was thinner than in the control group [ （0. 025 ± 0. 005 ）, （0. 093 ± 0. 024 ） ram, （ P 〈 0. 05 ） ]. However, there was no statistically significant differ- ence in the rate of cell proliferation between two groups （ P 〉 0. 05 ）. The apoptosis rate in the experimental group was higher than in the control group [（3.3±0.7）%, （6.7±1.5）% vs （1.2±0.4）%, （1.8± 0. 5 ） % I in 4 and 12 weeks after grafting （ P 〈 0. 05 ）. Conclusion Treatment of vein graft with glutaraldehyde can significantly reduce the number of graft smooth muscle cells and inhibit intimal hyperplasia.
