摘  要:目的观察镁锌合金(Mg-Zn)对小鼠前成骨细胞(MC3T3-E1)整合素表达的影响。方法将MC3T3-E1细胞以1.0×10^5个/ml密度接种于Mg-Zn和左旋聚乳酸(PLLA)上,共培养1、3、6、9、12和15d后,收集细胞,利用实时荧光定量聚合酶链反应(PCR)方法检测整合素亚基α2、α5和B1(Itgα2、Itgα5和Itgβ1)mRNA的表达水平。结果Itβ1mRNA表达水平最高,且随着时间的延长而增高(P〈0.01),但两组材料差异无统计学意义(P〉0.05);Mg-Zn表面Itgα5mRNA表达水平随着时间延长而增高(P〈0.01),除第1天外,PLLA表面Itgα5mRNA表达水平均低于Mg-Zn表面(P〈0.01);Itgα2mRNA表达水平随着时间延长而增高,在第9天时到达高峰,随后逐渐降低。在第1、3、6天Mg—Zn表面Itgα2mRNA表达水平高于PLLA(P〈0.01),而第9、12、15天时两组差异无统计学意义(P〉0.05);Mg-Zn表面Itgα2mRNA表达水平除了第15天外均高于Itgα5mRNA(P〈0.01)。结论与PLLA比较,Mg—Zn能够提高成骨细胞Itgα5mRNA和Itgα2mRNA表达水平,有利于促进成骨细胞与材料表面的黏附。Objective To observe the effect of Mg-Zn alloy on the integrin expression of preosteo- blasts MC3T3-E1 Methods The integrin α2, α5, β1 mRNA expression of 1 0 × 10^5/ml MC3T3-E1 euhured with MgZn and PLLA was detected by real time fluorescent quantitative polymerase chain reaction (PCR) after 1, 3, 6, 9, 12 and 15 days Results The mRNA expression of integfiu β1 in osteoblasts treated with Mg-Zn and PLLA was the most abundant and increased with time (P 〈 0 01 ), but there was no significant difference between the two groups ( P 〉 0 05 ) ; The mRNA expression of integTin α5 on Mg- Zn was increased with time ( P 〈 0 01 ), and that of integrin α5 on PLLA was totally lower than that on Mg-Zn except at the first day (P 〈0 01 ) ; The mRNA expression of α2 on Mg-Zn and PLLA was increased with time and reached the peak at 9th day with following reduction. The mRNA expression of integrin (α2 on Mg-Zn was higher at 1st, 3rd, 6th day than that on PLLA (P 〈0.01 ) and there was no significant difference between the two groups at 9th, 12th, 15th day ( P 〉 0. 05 ) ; The mRNA expression of integrin α2 was higher than integrin α5 on Mg-Zn except at the 15th day(P 〈0. 01 ). Conclusion Compared with PLLA, Mg-Zn enhanced the mRNA expression of integrin α2 and α5 of osteoblasts, which would benefit to the attachment of osteoblasts with the materials surface.
