摘 要:目的利用新型微流控装置建立稳定、线性的美洲大蠊提取物浓度梯度环境,观察美洲大蠊提取物对血管内皮细胞迁移的影响。方法将细胞接种于微流控装置,分别用培养基与提取液以体积比分别为80:20和95:5的混合液诱导内皮细胞6h,并于显微镜下观察血管内皮细胞在美洲大蠊提取物诱导下的迁移。结果当培养基与提取液体积比为80:20时,低浓度区血管内皮细胞分别减少7%和3%,高浓度区细胞分别增加9%和2%;当培养基与提取液体积比为95:5时,血管内皮细胞未出现明显的向高浓度迁移现象。结论美洲大蠊提取物能够诱导血管内皮细胞迁移。Objective We designed a new microfluidic device to generate stable,linear concentration gradients of extraction from Periplaneta americana in a cell culture chamber,in order to study migration of human vascular endothelial cells in response to quantified gradients. Methods The cells were inoculated to the microfluidic device, and then exposed to different concentration gradients of extraction ( Medium and extraction volume ratio 80: 20,95: 5) for 6 hours. Results The low concentration zone of vascular endothelial cells was decreased by 7% and 3% ,and high-concentration zone cells were increased by9% and 2% when the Medium and extraction volume ratio was 80 : 20. In contrast, shallow gradient ( Medium and Periplaneta americana volume ratio 95 : 5 ) were unable to induce human endothelial cells migration. Conclusion These results suggest extraction from Periplaneta americana could induce vascular endothelial cell migration.
