摘 要:目的:验证荧光分光光度法测定人体尿中维生素B2分析方法的准确性和稳定性,并对其在临床检验运用中的可行性、时效性进行探讨。方法:采用硅镁吸附柱对尿样进行净化除杂前处理,以分离提纯尿中维生素B2,并在荧光分光光度计激发波长445 nm,发射波长523 nm下测定,比较维生素B2标准液与尿样的荧光值强度,对尿中维生素B2含量进行测定。结果:方法的相对标准偏差(n=6)为1.44%,加标回收率在90%~100%之间,方法的线性范围为0~4.2μmol/L。结论:采用荧光分光光度法对尿中维生素B2含量进行分析测定,方法简便易行,基本满足临床检验要求。Objective: To verify the accuracy and stability of fluorescence spectroscopy for determination of the vitamin B2 in urine specimen,and to discuss the feasibility and the efficiency of the method.Methods: Si-Mg adsorption column was employed to purify and separate the vitamin B2 from the urine sample.At emission wavelength of 523 nm and excitation wavelength of 445 nm,the fluorescence of urine samples and standard samples were measured and the content of vitamin B2 in urine samples were calculated by compared the fluorescence of urine samples with standard sample.Results: The relative standard deviation of 6 samples is 1.44%,the rate of recovery ranges 90% to 100%,and the linear range is 0~4.2 μmol / L.Conclusion: The method of fluorescence spectroscopy for urinary vitamin B2 determination is accurate and feasible,and can meet the basic requirement of the clinical examination.
