摘 要:目的:对更昔洛韦超声定位靶向释药微囊的制备工艺、含量测定及体外释药行为进行初步研究。方法:用双乳化溶剂挥发法制备微囊,正交试验优化制备工艺,紫外分光光度法测定微囊包封率与载药量,超声诊断仪进行体外超声成像试验,高效液相色谱法测定微囊体外药物释放情况。结果:根据优化工艺制备的更昔洛韦微囊外型圆整,平均粒径为1.28μm,包封率为40.81%,载药量为3.02%,体外成像清晰,未加超声影响时微囊具有良好的缓释作用,在超声影响下微囊可在25min内将药物释放完毕。结论:更昔洛韦超声定位靶向释药微囊具有在超声控制下释放药物的能力,有潜在的应用价值。OBJECTIVE To study the preparation process, assay and in vitro release evaluation of ultrasonic locating and target-releasing microcapsule for Ganciclovir delivery. METHODS The microcapsules were prepared by compound emulsiondispersion solvent evaporation method and orthogonal design. The envelope rate and drug loading were detected by UV spectro photometer. Diagnostic image of microcapsules were watched in vitro. HPLC was used to study in vitro release of microcap sules. RESULTS The microcapsules were regular in their morphology; the average particle size was 1.28 um; the envelope rate was 40.81% and the drug loading was 3. 02%. The diagnostic image was clear. The microcapsules without ultrasonic showed significant sustained release ability without ultrasonic and they could release drug in 25 min with ultrasonic effect. CONCLUSION The ultrasonic locating and traget releasing microcapsules show control release ability. It may have potential appli cation value.
