摘 要:目的:研究骨癌痛模型大鼠脑脊液兴奋性氨基酸谷氨酸含量的变化。方法:雌性SD大鼠32只,随机分为4组,每组8只。Ⅰ为正常对照组;Ⅱ为D-Hank′s组;Ⅲ为热灭活细胞组:大鼠右侧胫骨上段骨髓腔注射热灭活的Walker256乳腺癌细胞10μL;Ⅳ为骨癌痛模型组。分别对各组大鼠行为学检测后,抽取大鼠脑脊液,应用反相高效液相色谱法,检测各组大鼠脑脊液内兴奋性氨基酸谷氨酸的含量。结果:Ⅳ组机械性痛觉缩爪阈值和热辐射痛觉缩爪阈值显著低于其他各组(P<0.05),而脑脊液中谷氨酸含量(19.41±0.64)μg/mL显著高于其他各组(P<0.05)。Ⅰ、Ⅱ和Ⅲ组之间大鼠脑脊液谷氨酸含量无统计学差异(P>0.05)。结论:中枢神经系统谷氨酸含量与骨癌痛的形成和维持密切相关。Objective To investigate the variation of glutamate concentration in cerebrospinal fluid(CSF) in a rat model of bone cancer pain. Methods Thirty-two female SD rats were randomly divided into 4 groups, 8 rats in each group:group Ⅰ , naive group; group Ⅱ , D-Hunk's group; group Ⅲ, heat-killed cells group; group Ⅳ, bone cancer pain group. After behavior tests, the glutamate concentrations in the rat's CSF were detected by reversed phase high performance liquid chromatography. Results The mechanical withdrawal threshold and thermal withdrawal latency were significantly decreased in group Ⅳ as compared with the other three groups (P 〈 0.05). Glutamate concentration in CSF of group Ⅳ (19.41 ± 0.54)μg/mL was significantly higher than the other three groups (P 〈 0.05). No marked differences were detected in the glutamate concentration in the rat's CSF among the group Ⅰ , Ⅱ,and Ⅲ (P 〉 0.05). Conclusions The glutamate concentration in CSF was significantly increased, which might play a role in the facilitation and maintenance of bone cancer pain.
