摘  要:目的:观察地龙水提取物对大鼠大脑中动脉缺血再灌注诱导凋亡的影响。方法:大鼠灌胃地龙水提取物(相当生药量1.4 g/kg)14 d后经大脑局灶性脑缺血再灌注损伤造模,运用HE组织染色、Caspase-3免疫组化染色、Hoechst 33342荧光染色及DNA琼脂糖电泳进行观察。结果:地龙水提取液能有效地减轻缺血再灌注损伤导致的大脑皮层水肿、充血,改善神经元的正常形态结构;地龙水提取液能显著地降低大脑皮层损伤后的caspase-3蛋白表达,降低神经元凋亡。结论:地龙水提取液有良好地抗大鼠大脑缺血再灌注诱导的神经元凋亡作用。Objective: To study the anti-apoptosis effects of earthworm-liquid on cerebral injury after focal ischemic reperfusion in rats. Methods: Before the animal model was established by middle cerebral artery occlusion(MCAO) , each rat was treated with earthwormliquid at dose 1.4g/kg by intragastric administration for 14 days. We investigated the anti-apoptosis effects of earthworm-liquid by H. E staining, easpase-3 immuno-histoehemieal staining, Hoechst 33342 staining and DNA agarose gel eleetrophoresis. Results: Earthworm-liquid can significantly reduce the cerebral cortex edemata and hyperemia which produced by MCAO, improve the normal structures of the neurons; The earthworm-liquid can significantly degrade the expressions of caspase-3 protein in the cerebral cortex after surgery injury, both decrease the neuron apoptosis. Conelusion: earthworm has the anti-apoptosis effect against cerebral isehemiareperfusion in rats.
