黄芪注射液对小鼠骨髓基质细胞黏附分子表达的影响

摘  要:目的:观察黄芪注射液对小鼠骨髓基质细胞表面细胞间黏附分子-1(ICAM-1)、血管细胞间黏附分子-1(VCAM-1)表达的影响。方法:(1)小鼠骨髓基质细胞体外培养;(2)倒置相差显微镜动态观察、HE染色光镜及透射电镜观察小鼠骨髓基质细胞的形态;(3)MTT法检测黄芪注射液对骨髓基质细胞增殖的影响;(4)采用流式细胞术检测黄芪注射液对小鼠骨髓基质细胞表面VCAM-1和ICAM-1的表达。结果:(1)MTT法检测黄芪注射液浓度为400μg/mL和600μg/mL均可促进小鼠骨髓基质细胞增殖(P<0.05),且两种剂量组间比较无差异;(2)与对照组比较,黄芪注射液能够增加小鼠骨髓基质细胞表面VCAM-1和ICAM-1蛋白表达(p<0.05)。结论:黄芪注射液能促进小鼠骨髓基质细胞的增殖及表面VCAM-1和ICAM-1蛋白的表达。提示黄芪注射液对造血微环境有改善作用。Objective:To investigate the effect of radix astragali injection on expression of ICAM-1 and VCAM-1 in bone marrow stromal cells(BMSCs) surface in mouse,and to approach the regulatory mechanism of radix astragali injection on haematogenesis.Methods:(1) Mouse BMSCs were cultivated in vitro.(2) Morphology of mouse BMSCs was idendified by inverted phase contrast microscope,hematoxylin and eosin(HE)staining and transmission electron microscope.(3)The concentration of radix astragali injection on promoting BMSCs proliferation was detected with MTT.(4) The expression of VCAM-1 and ICAM-1 in BMSCs was detected with flow cytometry.Results:(1) The results of MTT showed that the concentrations of radix astragali injection at 400μg /mL and 600μg /mL both promoted BMSCs proliferation(P0.05),and this two concentrations had no differences significantly within groups.(2) The expression levels of VCAM-1 and ICAM-1 in radix astragali group were higher than that of control group(P0.05).Conclusion:Radix astragali injection promoted BMSCs proliferation and the expression of VCAM-1 and ICAM-1 proteins in BMSCs.It is suggested that radix astragali injection could improve hematopoietic microenvironment.
