摘　　要：细胞周期及生长的调控是细胞存活必不可少的,在细胞增殖及细胞周期控制有许多复杂的机制。Gadd45a基因为生长阻滞和DNA损伤修复基因,是重要的抑癌基因,是p53、BRCI基因调控的下游基因,调控细胞周期阻滞,细胞纺锤体检查点调控,DNA修复和诱导细胞凋亡。在细胞对外界损伤反应调控网络中扮有重要角色,在维持基因组稳定性和抑制肿瘤发生发展的过程中发挥重要的作用。Gadd45a作为控制癌细胞增殖代谢途径的重要组成部分,成为一个值得新兴的药物进一步研究靶点。Regulation of cell cycle and growth is integral for cell survival.The intricate mechanisms that control proliferation and cell cycle are numerous.The tumor-suppressor gene Gadd45a is the most important anti-oncogene,Gadd45a is a p53-regulated downstream gene.It is known that p53 stimulates the promoter activities of Gadd45a gene to enhance their expression as a transcriptional factor,resulting in cell cycle arrest,cellular spindle checkpoint regulation,DNA repair and apoptosis,Gadd45a is a BRCA1-regulated downstream gene and mediates the role of BRCA1 in the control of cell cycle G2/M arrest and growth suppression.As Gadd45a is an essential component of many metabolic pathways that control proliferating cancer cells,it presents itself as an emerging drug target worthy of further investigation.
