摘 要:目的:研究健康人替硝唑血浆和唾液浓度的相关性,为临床用唾液浓度代替血液浓度提供依据.方法:8名男性健康志愿者分别po单剂量替硝唑片剂2.0g,采用高效液相色谱法(HPLC)测定替硝唑唾液和血浆浓度,并比较两种浓度各自计算的药物动力学参数.结果:替硝唑血浆浓度在1.13~44.9mg/L,唾液浓度(C_n)和血浆浓度(C_p)呈显著的正向相关关系(P<0.01).(C_n/C_p)为0.74±0.21(n=72).分别由C_n.和C_p,计算的药物动力学参数T_(1/2(K_a))、T_(1/2(k_e))、t_(peak)较为接近,无显著住差异(P>0.05),结论:可用唾液浓度代替血浆浓度进行替硝唑的药物浓度监测和人体药物动力学研究.AIM:To study correlation between plasma and saliva concentration of tinida-zole in healthy subjects. METHODS: A single oral dose of 2. 0 g tinidazole tablet was given to 8 healthy male volunteers respectively. The drug concentrations in plasma and saliva were determined by HPLC method. The pharmacokinetic parameters calculated by 2 concentrations respectively were compared. RESULTS:The linear regression equation of tinidazole salivary (c_ ) and plasma (cp) concentration were c_ = 0. 727 9 c_p - 0. 1322, r = 0. 9409, there was a significant positive correlation between c_ and c_p when its plasma concentration is 1. 13-44. 9 mg/L. The ratio of c_ /c_p was 0. 74±0. 21(n = 72). The pharmacokinetic parameters (T_1/2(k_4)), T_(1/2(k_4)), t_(peak) calculated by c_ and c_p respectively were near,there were no significant differences between them(P>0. 05). CONCLUSION:The drug concentration can be monitored and its pharmacokinetics in human body can be studied by the saliva concentration replacing the plasma concentration.
