摘 要:目的研究白血病抑制因子(leukemia inhibitory factor,LIF)对肾间质成纤维细胞活化的干预作用。方法体外培养正常大鼠肾脏成纤维细胞,分对照组、转化生长因子β1(transforming growth factor-β1,TGF-β1)处理组及TGF-β1和LIF共处理组,电镜下观察细胞形态变化,通过RT-PCR及Western blot检测肌成纤维细胞标志物α-平滑肌肌动蛋白(alpha-smooth muscle actin,α-SMA)表达的改变,双抗体夹心ABC-ELISA检测细胞上清Ⅰ型胶原。结果 TGF-β1引起成纤维细胞激活,产生形态学改变,α-SMA表达增高(P<0.01),Ⅰ型胶原产生增加(P<0.01)。与TGF-β1处理组相比,LIF干预组形态学改变不明显,LIF能干预TGF-β1引起大鼠肾间质成纤维细胞α-SMA表达和Ⅰ型胶原产生。结论 LIF可以抑制TGF-β1引起的肾间质成纤维细胞激活。Objective To investigate the effects of leukemia inhibitory factor(LIF) on renal interstitial fibroblast activation following induction by transforming growth factor beta 1(TGF-β1).Methods Normal rat interstitial fibroblast cells(NRK/49F) were treated with TGF-β1 and TGF-β1 combining with LIF respectively for different duration with different concentration.Changes in cell morphology and expression of α-SMA were evaluated with electronic microscope and Western blot respectively.The collagen Ⅰ in the supernatant was detected with ABC-ELISA.Results TGF-β1 induced renal interstitial fibroblast activation,and this was accompanied by significant morphological transformations and secretion of collagen Ⅰ.Co-culturing of cells with LIF blocked the morphological transformation.In addition,LIF inhibited TGF-β1-induced expression of α-SMA mRNA and protein(P〈0.01),and decreased the levels of collagen Ⅰ(P〈0.01) in a dose-dependent manner.Conclusion LIF suppresses TGF-β1-induced activation and collagen Ⅰ secretion of cultured renal interstitial fibroblasts.
