摘  要:目的探讨郁可舒胶囊对脑卒中后抑郁(PSD)模型大鼠脑内单胺类递质的影响及中医药治疗PSD的机理。方法大鼠随机分为正常对照组、模型组、假手术组、百优解组、郁可舒胶囊高、中、低剂量组,采用颈内动脉线栓法制备局灶性脑缺血大鼠模型并予以行为限制制备PSD大鼠模型,在造模29d后采用荧光检测法测定大鼠左、右侧额顶皮层和脑干区单胺类递质的变化。结果复合造模处理可引起大鼠脑内5-羟色胺(5-HT)、去甲肾上腺素(NE)、多巴胺(DA)显著降低,百优解和郁可舒胶囊均能显著增加PSD大鼠脑内5-HT、NE、DA水平。结论郁可舒胶囊可以通过提高PSD大鼠的脑内单胺类递质水平而起到治疗作用。Objective:To observe the effect of Yukeshu Capsule on monoamine transmitters in cerebral of post-stroke depression (PSD) rats and to detect the mechanism of TCM on PSD.Methods:Rats were divided randomly into the normal group,the model group,the sham-operated group,the fluoxetine group,the high dose of Yukeshu Capsule group,the middle dose of Yukeshu Capsule group and the low dose of Yukeshu Capsule group.After being given the suture method in the internal carotid artery,the PSD rat model was established by the behavior restriction.In the 29th day after model establishment,the left and the right frontoparietal cortex and the monoamine transmitter in brainstem were detected by the fluorescence assay.Results:These levels of 5-HT,NE and DA were decreased obviously.These concentrations were increased by the fluoxetine or the Yukeshu Capsule.Conclusion:Yukeshu Capsule could treat PSD by increasing the level of monoamine transmitters in rats’cerebral.
