摘  要:目的探寻中药复方清热颗粒对耐环丙沙星铜绿假单胞菌外排泵影响的作用机制。方法建立急性耐环丙沙星铜绿假单胞菌腹腔感染兔动物模型,复方清热颗粒药液灌胃,每日2次,连续5d。末次给药后60min无菌心脏取血,离心分离出含药血清。应用外排泵抑制剂(CCCP)选择出外排泵阳性的耐环丙沙星铜绿假单胞菌9株,应用复方清热颗粒含药血清和环丙沙星(CIP)进行干预,将干预前后的铜绿假单胞菌分别行增菌培养,提取外膜蛋白,行蛋白电泳,紫外凝胶成像后分析外膜蛋白中OprM量的变化来推测复方清热颗粒对铜绿假单胞菌外排泵的作用机制。结果复方清热颗粒含药血清作用前后铜绿假单胞菌外膜蛋白O-prM量的变化相近。结论复方清热颗粒含药血清可抑制铜绿假单胞菌的外排泵,但不能改变其外排泵的含量。Objective:To observe the mechanism of the compound clearing-heat granules on the efflux pump of the CIP-resistance pseudomonas aeruginosa.Methods:Rabbit models were established by being infected from the CIP-resistant pseudomonas aeruginosa in abdominal.These models were taken compound clearing-heat granules twice a day.This therapy was lasted five days.Sixty minutes after the last medicine,the drug serums were separated from heart blood.9 mutants of positive efflux pump CIP-resistance pseudomonas aeruginosa were selected by CCCP.After being taken the compound clearing-heat granules and the CIP,the pseudomonas aeruginosa were cultured and extracted outer membrane protein.The changes of the OprM,being detected by the protein electrophoresis and the uv-gel imagine,were used to presume the mechanism of the compound clearing-heat granules on the efflux pump of the CIP-resistance pseudomonas aeruginos.Results:There was no significant difference before and after being used the compound clearing-heat granules in the concentration of OprM of pseudomonas aeruginosa.Conclusion:Compound clearing-heat granules can inhibit the efflux pumps of the peudomonas aeruginosa.However,it could not change the content of efflux pumps of it.
