摘 要:目的:建立白屈菜药材的UPLC-UV特征图谱并用QTOF-MS技术分析其生物碱类成分。方法:采用ACQUITY UPLC BEH C18(2.1mm×100mm,1.7μm)色谱柱,以乙腈-10mmol.L-1醋酸铵缓冲液(用醋酸调节至pH3.0)为流动相梯度洗脱,流速0.4mL.min-1,检测波长280nm,柱温35℃,对10批白屈菜药材的特征图谱进行比较研究,用UPLC-ESI-QTOF-MS技术对其共有峰进行分析。结果:获得白屈菜药材的UPLC特征图谱,共有峰17个,鉴定出14个生物碱成分,其中6个通过对照品对照确认,8个通过质谱信息推测。结论:建立的UPLC-UV特征图谱可用于白屈菜质量评价。Objective:To establish the UPLC specific chromatogram of Chelidonium majus L.and analyze the alkaloids compositions by QTOF-MS.Methods:The samples were separated by ACQUITY UPLC BEH C18 column(2.1 mm×100 mm,1.7 μm)and eluted with acetonitrile-10 mmol·L-1 ammonium acetate buffer(adjusted to pH 3.0 with acetic acid)at a flow rate of 0.4 mL·min-1.The detection wavelength was 280 nm and column temperature was 35 ℃.In the condition,10 samples of Chelidonium majus were analyzed.Common peaks were analyzed by UPLC-ESI-QTOF-MS.Results:The specific chromatograms of Chelidonium majus L.were obtained.There were 17 common peaks.Fourteen alkaloids compositions were identified.Among them,6 compositions were identified by direct comparison with the reference substances and others were identified by MS and MS2 data.Conclusion:UPLC specific chromatogram facilitates the evaluation of the quality of Chelidonium majus L..
