摘  要:目的:建立RP-HPLC法同时测定不同药用部位、不同产地、不同季节的旱芹中绿原酸和咖啡酸的含量。方法:采用Kromasil C18柱(5μm,250mm×4.6mm),流动相为甲醇-0.05%磷酸水溶液梯度洗脱,流速1.0mL.min-1,检测波长327nm,柱温35℃。结果:绿原酸和咖啡酸进样浓度分别在3.71~92.8μg.mL-1(r=0.9999)和0.148~3.70μg.mL-1(r=0.9997)范围内与峰面积呈良好的线性关系,平均加样回收率(n=9)分别为100.3%和100.2%。结论:分析方法快速、准确,重现性好,可用于控制旱芹的质量。Objective:To establish an RP-HPLC method for the simultaneous determination of chlorogenic acid and caffeic acid in Apium graveolens L.of different parts,different habitats and different seasons.Methods:The chromatographic method was carried out on Kromasil C18 column(5 μm,250 mm×4.6 mm)and methanol-0.05% phosphoric acid with gradient elution as the mobile phase was adopted;The flow rate was 1.0 mL min-1;The absorbance was monitored at 327 nm,and the column temperature was 35 ℃.Results:The linear response ranges were 3.71-92.8 μg·mL-1(r=0.9999)and 0.148-3.70 μg·mL-1(r=0.9997),respectively.The average recoveries(n=9)were 100.3% and 100.2%,respectively.Conclusion:The assay demonstrates that the method has adequate accuracy and selectivity to measure the concentration of chlorogenic acid and caffeic acid in Apium graveolens L.
