南葶苈子药材含量标准的研究

摘　　要：目的:建立高效液相色谱法测定南葶苈子药材中槲皮素-3-O-β-D-葡萄糖-7-O-β-D-龙胆双糖苷的含量,确定南葶苈子药材的含量标准。方法:供试品溶液制备方法为70%甲醇回流1h,色谱柱为VenusilMP C18(250mm×4.6mm,5μm),流动相为乙腈-0.1%醋酸水溶液(11:89),流速1.0mL.min-1,检测波长254nm,柱温25℃。结果:槲皮素-3-O-β-D-葡萄糖-7-O-β-D-龙胆双糖苷进样量在0.156～1.56μg范围内与峰面积呈良好的线性关系;加样回收率为99.12%,RSD=1.3%(n=6)。结论:本方法分离度较好,具有很好的重现性和稳定性。综合28批南葶苈子药材含量测定结果,槲皮素-3-O-β-D-葡萄糖-7-O-β-D-龙胆双糖苷的含量限度应不低于0.075%。Objective：To establish a method to determine the content of quercetin-3-O-β-D-glucopyranosyl-7-O-β-D-gentiobioside in Semen Descurainiae,and to make the chemical content standard of the herb.Methods：The method of sample solution preparing was refluxing the herb in 70% methanol for 1 h.In HPLC method,the self-made quercetin-3-O-β-D-glucopyranosyl-7-O-β-D-gentiobioside was used as reference substances.The chromatographic conditions were as follows：MP C18 column（250 mm×4.6 mm,5 μm）,acetonitrile-0.1% acetic acid solution（11：89）as mobile phase,the follow rate 1.0 mL·min-1,detection wavelength at 254 nm,the temperature of column 25 ℃.Results：The linear range was 0.156-1.56 μg for the reference compound.The average recovery was 99.12%with RSD of 1.3%（n=6）.Conclusion：The method is simple and accurate,with good separation,repeatability and stability.Through the results of 28 batches of the herb,the content of quercetin-3-O-β-D-glucopyranosyl-7-O-β-D-gentiobioside in Semen Descurainiae should not be less than 0.075%.
