摘  要:目的:对苍术样品进行定性、定量试验研究,提高苍术的质量标准。方法:运用薄层色谱法对苍术中聚乙炔类成分苍术素、苍术素醇和(4E,6E,12E)-十四癸三烯-8,10-二炔-1,3-二乙酸酯进行定性分析;采用高效液相色谱法测定苍术素的含量,色谱柱为Ultimate AQ-C18(4.6mm×250mm,5μm)柱,流动相为甲醇-水(79:21),流速1.0mL.min-1,检测波长340nm,柱温30℃。结果:苍术的薄层色谱鉴别特征明显,专属性强;苍术素进样量在0.0018~0.63μg范围内与峰面积呈良好的线性(r=1.000),回收率>97.5%(RSD<1%,n=3)。结论:通过31批不同来源苍术药材、苍术饮片、麸炒苍术的鉴别和含量测定,证明方法灵敏,准确可靠,重复性好。可用于苍术的质量控制,具有很高的实用价值。Objective:To establish qualitative and quantitative methods for the quality elevation of Rhizoma Atractylodis.Methods:Three polyacetylene components of atractylodin,atractylodinol and (4E,6E,12E)-tetradecatriene-8,10-diyne-1,3-diyl diacetate were identified by TLC.The separation of atractylodin was obtained by RP-HPLC using Ultimate AQ-C18(4.6 mm×250 mm,5 μm) and elution systems of methanol-water (79:21) with a flow rate of 1.0 mL·min-1 at 30 ℃;The detection wavelength was at 340 nm.Results:The identification of TLC was characteristic and specific.Calibration curve showed good linear regression(r=1.000) within test ranges of 0.0018-0.63 μg.Average recovery was greater than 97.5%(RSD1%,n=3).Conclusion:The developed methods were proved to be sensitive,reliable and useful by analysis of thirty-one different samples from Rhizoma Atractylodis,pieces of Rhizoma Atractylodis and bran-processed of Rhizoma Atractylodis.This rapid and accurate method has been successfully applied to the quality evaluation of Rhizoma Atractylodis.
