摘  要:目的考察G-2407-3鼻腔喷雾剂及其辅料的鼻黏膜毒性作用。方法采用离体和在体蟾蜍上腭模型法,光学显微镜下观察纤毛持续运动时间,考察辅料和G-2407-3的纤毛毒性作用。结果离体法考察辅料A、B、三氯叔丁醇、羟苯乙酯对纤毛运动有影响,但是影响程度不同;通过在体法考察G-2407-3鼻腔喷雾剂对纤毛运动的影响,结果显示影响较少,离体法考察辅料B、羟苯乙酯对纤毛有一定的毒性,而辅料A、三氯叔丁醇毒性较小,而且均可逆。不同浓度的G-2407-3对纤毛有一定的毒性,该实验剂量下这种影响是可逆的。pH值在4~8范围内对纤毛运动影响较小,其中pH值为6.0时对纤毛影响最小;在体法考察G-2407-3与上述结果基本一致。结论G-2407-3鼻腔喷雾剂具有可行性。Objective To investigate the nasal toxicity of G-2407-3 nasal sprays and excipients.Methods In the experiment,toad palate models were used in vitro and in vivo.Ciliary toxicity of excipients and G-2407-3 was investigated and evaluated by the duration of ciliary movement.Results Auxiliary B and nipagin were toxic in vitro;auxiliary A and acetone chloroform were less toxic,and the toxicity was reversible.Different concentrations of G-2407-3 all had ciliary toxic while the effect of experimental doses was reversible.The pH value from 4 to 8 had less effect on ciliary movement,among which pH value of 6.0 had the minimal effect.G-2407-3 was basically consistent with the above results in vivo.Conclusion G-2407-3 nasal spray is feasible.
