摘  要:目的对中药复方海藻制剂水煎液水相的化学组成进行初步研究。方法采用苯酚-硫酸法测定复方海藻制剂水煎液水相以及水相醇沉粗多糖的总糖含量;采用硫酸钡-明胶比浊法测定两者的硫酸基含量;采用硫酸咔唑法测定两者的糖醛酸含量;采用Folin酚法测定两者的蛋白质含量。采用气相色谱法对两者的单糖组成进行分析。结果复方海藻制剂水相总糖含量为56.90%,硫酸基含量为25.71%,糖醛酸含量为6.32%,总蛋白含量为38.01%。单糖组成分析显示水相中中性糖主要为葡萄糖,还有少量岩藻糖,甘露糖,半乳糖以及微量的鼠李糖。醇沉后粗多糖含量为53.05%,硫酸基含量为32.24%,糖醛酸含量为7.74%,总蛋白含量为22.01%。单糖组成分析显示粗多糖中性糖主要以葡萄糖为主。结论水相的化学成分主要是以由葡萄糖为主要中性糖组成的多糖类成分,而多糖可能是复方抗肿瘤重要的药效成分。Abstract: Objective To study the chemical composition of the water fraction from the traditional Chinese medicine Seaweed Compound Preparation (SCP). Methods The contents of polysaccharides of the water fraction and the crude polysaccharide from SCP were determined by phenol-sulfuric acid method. The contents of sulfate radical and glucuronic acid were evaluated by barium sulfate-gelatin method and sul- furic acid carbazole colorimetry method, respectively. The protein contents were measured by the Folin phenol method. The monosaeehride composition was determined by the gas chromatography method. Results The content of polysaccharides of the water fraction was 56. 90%, the sulfate radical content was 25. 71%, the glucuronic acid content was 6.32% and protein content was 38. 29%, respectively.Monosaccharide analysis showed that higher content neutral monosaccharide of glucose with small a- mounts of fucose, mannose, galactose and rhamnose were in the water fraction. The content of poly- saccharides of the crude polysaccharide from SCP was 53. 05%, the sulfate radical content was 32. 24%, the glucuronic acid content was 7. 74% and protein content was 22. 01%, respectively. High content of glucose was found in crude polysaccharides from the monosacchride composition analysis. Conchtsion The main chemical constituent of the water fraction was polysaccharide which was almost constituted by glucose. The polysaccharide might be the important pharmaceutical effective constituents of SCP.
