摘 要:目的优化缘管浒苔多糖提取分离工艺,获得最佳工艺。方法采用热水浸提法设计单因素试验及4因素3水平正交试验,对缘管浒苔多糖提取工艺进行了优化,采用乙醇沉淀法对缘管浒苔多糖的分离工艺进行研究。结果4因素对缘管浒苔多糖得率的影响因素顺序为料液比>提取次数>提取时间>提取温度,提取最佳工艺为:料液比1:20,提取时间5h,提取温度85℃,提取次数3次。最佳分离条件为3倍体积的95%乙醇沉淀6h。结论优化后的缘管浒苔多糖提取分离工艺稳定可行,在该条件下,缘管浒苔多糖的得率为5.37%。Objective To optimaize the methods of extraction and separation technologies of Enteromorpha linza polysaccharides. Methods Single factor tests and orthogonal experiments were carried out hy using four factors and three levels design, for the Enterornorpha linza poIysaccharide extraction technology by water bath. The separation experiment was carried out by using 95% ethanol in different volumes and precipitation time. Results The four influent factors of water hath were in order: ratio of solid to liquid〉extract frequency〉extract time〉extract temperature. The optimal extraction conditions obtained were: ratio of solid to liquid was 1 : 20, extract at 85℃, 5 hours for three times. The best conditions to precipitate polysaeeharide were by using three times volume of 95% ethanol solution for six hours. ConclusionThe optimum technology is easy to operate and stability, under these conditions the yield of polysaccharide was 5.37%.
