摘 要:①目的观察自拟中药复方益肾蠲湿合剂对肾小球系膜细胞(glomerular mesangial cell,GMC)分泌细胞外基质的抑制作用。②方法将1×10-3、1×10-2和1×10-1mg/mL益肾蠲湿合剂的提取液作用于含10%血清DMEM培养的GMC,分别于24、48和72h后收集培养上清,采用Elisa法检测上清中透明质酸(hyaluronic acid,HA)、纤维连结蛋白(FN)及Ⅳ型胶原(ColⅣ)含量。③结果 10%血清促进了GMC分泌HA、FN和ColIV,但这种作用可明显被不同浓度的益肾蠲湿合剂所抑制,并且其抑制程度随药物浓度的增加而增加。④结论益肾蠲湿合剂可以抑制体外培养的GMC分泌HA、FN和ColIV,推测这种作用是其治疗慢性肾小球疾病的主要机制之一。Objective To observe the inhibitory effects of Yishenjuanshi traditional Chinese herbs on the secretion of extracelluar matrix in cultured glomerular mesangial cells(GMCs).Methods GMCs cultured in 10% serum-DMEM were treated with 1×10^-3,1×10^-2 and 1×10^-1 mg/mL Yishenjuanshi Chinese herbs for 24,48 and 72 hours,then the supermatants were collected to detect the contents of hyaluronic acid(HA),fibronectin(FN) and collagen IV(ColⅣ) using ELISA method.Results The secretion of HA,FN and ColⅣ in GMCs were promoted by 10% serum,while which were significantly inhibited by different doses of Yishenjuanshi,and the inhibitory degrees increased as the doses of Yishenjuanshi increased.Conclusion The secretion of HA,FN and ColⅣ in GMCs could be inhibited by Yishenjuansh,which invlolved in the mechanisms of "Yishenjuanshi" inhibiting the development of chronic glomerulus diseases.
