盐酸小檗碱抑制人颊黏膜成纤维细胞炎症反应的初步实验研究

摘　　要：目的观察盐酸小檗碱对脂多糖刺激人颊黏膜成纤维细胞炎症反应的抑制作用。方法原代培养人颊黏膜成纤维细胞作为对照组;脂多糖刺激组作为炎症组,同时加入小檗碱和脂多糖组作为实验组。采用酶联免疫吸附测定(ELISA)法检测细胞培养上清液中前列腺素E2的含量作为细胞炎症反应指标。结果炎症组较对照组上清液中前列腺素E2浓度明显升高[(8.682±1.984)μg/L vs(22.509±1.245)μg/L,P<0.05],药物组较炎症组上清液中前列腺素E2浓度明显降低[(11.381±1.652)μg/L vs(22.509±1.245)μg/L,P<0.05]。结论盐酸小檗碱可以明显抑制由脂多糖刺激人颊黏膜成纤维细胞分泌的前列腺素E2。盐酸小檗碱在抗人颊黏膜成纤维细胞的炎症反应中具有重要的作用。Objective To explore the depressant effect of inflammatory reaction in human buccal mucosa fibroblasts（HBMFs） by berberine hydrochloride（Ber）.Methods To primarily culture HBMFs.HBMFs cultured in normal way was as the control group;HBMFs excited by lipopolysaccharide（LPS） as the inflammatory group;HBMFs excited by lipopolysaccharide（LPS） and Ber as the experimental group.To detect the density of prostaglandin E2（PGE2）in clear supernatant liquid of HBMFs with the method of enzyme linked immunosorbent assay（ELISA）.Results The density of PGE2 in the clear supernatant liquid of HBMFs in the inflammatory group advanced obviously than that in the control group,with statistical significance[（8.682±1.984）μg/L vs（22.509±1.245）μg/L,P0.05].The density of PGE2 in the clear supernatant liquid of HBMFs in the experimental group advanced obviously than that in the inflammatory group,with statistical significance[（8.682±1.984）μg/L vs（22.509±1.245）μg/L,P0.05].Conclusion Ber can obviously depress the density of PGE2 in HBMFs excited by LPS.Ber has the important effect in the anti-inflammatory reaction in HBMFs.
