三种治法方药对肝癌H_(22)荷瘤小鼠免疫功能影响的比较研究

摘　　要：目的:观察比较活血化瘀方、清热祛湿方和养阴柔肝方三种不同治法中药复方对H22荷瘤小鼠免疫功能的影响。方法:采用皮下接种肝癌H22细胞的方法建立荷瘤小鼠模型,分别用上述三种中药复方水煎液灌胃处理11d,称量各组脾脏重量,计算脾指数;应用双抗体夹心ABC-ELISA法检测荷瘤小鼠血清IL-2、TNF-α含量的变化。结果:活血化瘀、清热祛湿、养阴柔肝方对H22小鼠脾指数无显著影响(P>0.05);与正常对照组相比,模型对照组血清IL-2的含量显著降低(P<0.05),各用药组与模型对照比较显著升高(P<0.05),三个用药组之间两两比较,也具有显著性差异(P<0.05);与正常对照组相比,各组血清TNF-α的含量均显著升高(P<0.05),与模型对照比较,各用药组显著降低(P<0.05)。结论:三种治法均可有效提高移植性H22荷瘤小鼠的IL-2水平,提高小鼠的免疫力,从而对机体起到保护作用,这可能是其有效抗肝癌的疗效机制之一;三种治法(尤其是清热祛湿组)具有抑制荷瘤鼠机体TNF-α水平异常升高的作用,使其维持在一定水平而发挥抗肿瘤、调节机体免疫功能等积极作用。Objective：To observe the immune function of blood-activating and stasis -dissolving prescription,heatclearing and dampclearing prescription and nourishing-Yin and softeningliver prescription on H22 liver tumorbearing mice. Methods：The Hepatoma-bearing Mice model is made by subcutaneous injecting H22 cell,continuous gavage the water solution of those three prescription 11 days separately,Weigh each group of spleen weight,spleen index calculated; Using double-antibody sandwich ABC-ELISA method to test the level of IL-2 and TNF-αin serum of Hepatoma-bearing Mice. Results：Three prescription had no significant effect on spleen index of the H22 liver tumor-bearing mice（ P 0. 05） ; Compared with the normal group,the serum levels of IL -2 were significantly lower（ P 0. 05） ,and compared with the model group,the groups used three formulas was significantly higher （ P 0. 05） ,there also have significant differences between the three treatment group （ P 0. 05） ; Compared with the normal group,the serum levels of TNF-α were significantly higher（ P 0. 05） ,and compared with the model group,the groups used three formulas was significantly lower（ P 0. 05）. Conclusions：Three kinds of treatment methods can effectively improve the levels of IL -2 on H22 liver tumor-bearing mice,improve immunity in mice,thus play a protective effect on the body,this may be the effective mechanism of antiliver cancer; Three prescription（ Especially heat-clearing and dampclearing prescription） can inhibition the TNF-α levels of the body abnormal increased of H22 liver tumor-bearing mice,to maintain a certain level and play positive role at anticancer,modulation of immune function.
