摘  要:目的比较利多卡因和维拉帕米对乳腺癌细胞抗肿瘤药物的增敏作用。方法应用微量细胞培养四氮唑(MTT)法检测乳腺癌细胞系MCF-7对三种抗癌药物5-FU、MMC和CDDP敏感性。观察利多卡因和维拉帕米对三种化疗药物的增敏作用,并比较两药增敏作用之差异。结果 MCF-7对三种化疗药物均敏感,并随药物浓度的升高存活率降低。利多卡因和维拉帕米与三种化疗药物合用后分别降低三种化疗药物的半数抑制浓度(IC50),利多卡因比维拉帕米增敏作用强(P<0.01),加入利多卡因的增效倍数分别为27.4、30.1、59.1,而加入维拉帕米的增效倍数分别6.09、11.2、5.47。结论 MCF-7对三种化疗药物(5-FU、MMC及CDDP)均敏感,利多卡因和维拉帕米对三种化疗药物均有增敏作用,利多卡因比维拉帕米的增敏作用强。Objective To explore the difference between the increased sensitivity effect of Lidocaine and that of Verapamil. Methods By using MTF method,the sensitivity of the three anti-cancer drugs including 5-Fu, MMC and CDDP on the breast tumor cell line MCF-7 and the increased sensitivity of the three anti-cancer drugs by Lidocaine were studied. The difference between the increased sensitivity effect of Lidocaine and that of Verapamil were compared. Results MCF-7 was sensitive to all three anti-cancer drugs. The sensitivity increased gradually when concentration of anti-cancer drugs increased. The IC50 decreased when the three drugs combined with Lidocaine and Verapamil. There was significant difference in the increased sensitivity effect between Lidocaine and Verapamil(P 〈 0.01 ). The intensification factor of Lidocaine on the three anti-cancer drugs were 27.4, 30.1,59.1 respectively ( P 〈 0.01 ) and that of Verapamil were 6.09,11.2,5.47 respectively. Conclusion MCF-7 was sensitive to all three anti-cancer drugs. Lidocaine and Verapamil had increased sensitivity to three anti-cancer drugs. There was significant difference in the increased sensitivity effect between Lidocaine and Verapamil.
