摘  要:目的探讨经股静脉移植的大鼠真皮多能干细胞在缺血性损伤脑组织中的存活、迁移及分布情况。方法分离培养人真皮来源的多能干细胞(skin-derived precursors,SKPs),采用线栓法制作大鼠局灶性脑缺血再灌注模型,缺血2h后行再灌注,于再灌注24h后将CM-DiI标记的人真皮来源的SKPs经大鼠股静脉输入动物模型体内,植入后分别在第1、3、5d处死大鼠,作冰冻切片,采用免疫组织荧光染色法,检测SKPs在脑缺血大鼠体内的存活、迁移及分布情况。结果在移植了SKPs后的MCAO大鼠脑组织冰冻切片中,荧光显微镜下可见移植的SKPs。静脉移植的细胞主要分布在缺血灶的周围,健侧脑组织中偶见移植细胞。结论经股静脉移植的SKPs,能够在宿主缺血的脑组织中存活,并向损伤部位迁移。Objective To investigate the survival,distribution and migration of transplanted human skin-derived precursors in the ischemic brain in vivo.Methods Skin-derived precursors(SKPs)from human dermis was isolated.The right middle cerebral artery occlusion(MCAO)model of rat was established.The MCAO was obstructed using a single strand nylon thread and reperfused 2 hours later.SKPs labeled by CM-DiI were trans-planted into the right femoral vein of MCAO rat 24 hours after reperfusion.Fluorescent microscope was used to observe the survival,distribution and migration of transplanted SKPs.Results Transplated SKPs were observed in the brain frozen sections of MCAO rat which was transplanted with labeled SKPs:Most of transplanted cells mainly distributed around ischemic focus.There were few transplanted SKPs distributed in opposite hemicere-brum.Conclusion SKPs can be able to survive and migrate to ischemic brain tissue through intravenous trans-plantation.
