摘 要:目的研究胚胎干细胞(human embryonic stem cells,HESC)向神经祖细胞分化时,拟胚体(embryoidbody,EB)大小和碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)对诱导效率的作用。方法采用人胚胎干细胞形成的EB,按照其直径大小分为3组,拟胚体阶段添加或不添加bFGF分组,检测各组的贴壁效率和神经诱导效率。结果直径350~251μm的EB组和EB阶段未添加bFGF组的贴壁率与神经诱导效率高于其他组EB。结论优化EB大小和培养基成分可促进EB的神经诱导分化。Objective To study the influence of the size of embryoid body(EB) and basic fi-broblast growth factor(bFGF) on neural differentiation of human embryonic stem cells(HESC).Method EB derived HESC were divided into 3 groups according to their diameters,and grouped with or without bFGF addition during EB stage,then,the replating efficiency and neural induc-tion efficiency in all groups were detected.Results EB of the middle size group(d=350-251 μm) and without bFGF addition group had the highest replating efficiency and induction efficiency.Conclusion Optimization of EB size and medium component could help promote neu-ral differentiation.
