摘  要:目的研究蛇毒的抗肿瘤组分X(FX)的一般毒理学。方法分别给小鼠及家兔静脉注射五步蛇毒FX,采用King改良的Ruitman法、二乙醇-肟法等技术,观察FX急性毒性、长期毒性、出血毒性、神经毒性的影响。结果急性毒性实验中,FX的半数致死量为5mg/20g体重,属于毒性极低物质;长期毒性实验中,给药剂量为10mg/kg体重时,动物体重出现明显下降,但实验动物的生活状态、肾功能、肝功能、血常规检验均未见统计学上的差别;在出血毒性、神经毒性及过敏试验中,实验组均呈阴性反应。结论 FX应用于小鼠与对照组相比,其急性毒性、长期毒性、出血毒性、神经毒性无统计学差异(P>0.05),提示FX应用于临床也可能是安全的。Objective To examine the venom of anti-tumor component X(FX) in general toxicology.Method Mice and rabbits were given intravenous injection of the agkistrodon acutus venom componets X,King's improved Ruitman's law,di-ethanol-oximes-technologies were used in observation on influences of FX on acute toxicity,long-term toxicity,bleeding toxicity,neurotoxicity.Result In acute toxicity experiments,half of the lethal dose for 5 mg/20 g body mass,thus FX was extremely toxic substances;in long-term toxicity experiments,the dose of 10 mg/kg body weight,animal weight became a notable decline,but the living conditions of animals and their renal function,liver function,as well blood routine examination were of no statistically significant changes;in the bleeding toxicity,neurotoxicity,and allergy tests,the experimental group were all negative.Conclusion FX used in mice confirmed that compared with the control group,its acute toxicity,long-term toxicity,bleeding toxicity,neurotoxicity there was no statistical difference(P0.05),FX applications and clinical tips could also be safe.
