摘 要:本试验研究了不同浓度及组合消化酶对奶牛乳腺组织分离的影响以及不同FBS浓度培养基对奶牛乳腺上皮细胞纯化和生长的影响。试验结果表明0.25%胰酶+0.2%Ⅱ胶原酶对组织块分离效果最好,乳腺上皮细胞生长最多,5%与10%浓度FBS对奶牛乳腺上皮细胞纯化效果最好,5%与10%浓度FBS对细胞生长没有明显的差异(p>0.05),但明显高于其他处理组(p<0.01)。This experiment contains two parts.The purpose of the first part is to test the effect of different concentration and combination of digestive enzymes on the separation of breast tissue.The purpose of the second part is to test the effect of different FBS concentrations media on bovine mammary epithelial cells culture.The result indicate that accumulation of bovine mammary epithelial cells in treatment 0.25%Trypsin +0.2% collagenaseⅡwas highest among the other treatments,the bovine mammary epithelial cells cultured in media contain 5% and 10% concentration FBS were better purified than other treatments,and the bovine mammary epithelial cells number cultured in media contain 5% and 10% concentration FBS were have no significant difference(p0.05),but much higher than other treatments(p0.01).
