摘  要:为研究鸡源大肠杆菌分子流行病学规律,选用40株鸡源大肠杆菌(均含有质粒),应用指纹法对其质粒及染色体DNA进行了分析。对菌株的质粒图谱比较表明,来源相同的菌株,其图谱相同或相似;来源不同的菌株,其图谱一般不同。选取HindⅢ对质粒DNA进行酶切,进一步证实了质粒图谱分析的结果。对菌株质粒型(plasmidtype,PT)进行数值聚类分析,得出PTs间的聚类树状图,更直观地反映出菌株间的遗传关系。试验中还发现,多数鸡源大肠杆菌含有大质粒,它们可能含有毒力相关基因。选用HindⅢ、EcoRI分别对染色体DNA进行酶切,均能够产生较易于识别的酶切图谱。The DNA analysis of plasmids and its chromosomes for 40 strains of E.coli were performed by fingerprinting. The comparision of plasmids among strains demonstrated that the strains derived from the same source exhibited the identical or similar profiles and those from different sources exhibited the distinct profiles. Plasmid′s DNA cleaved with HindⅢ, further proved this result. The tree figure of the plasmid type(PT) was obtained by using the maximum linkage clustering method which would be more directly indicative of their genetic relations. There are some of the large plasmids in most of the E.coli strain which may exist the virulence related sequences. The chromosomal DNA, was digested with HindⅢ and Eco RI and the restriction profiles obtained in accordance with the sources of strains.
