摘 要:目的:观察补肾调经方(熟地、枸杞、覆盆子、淫羊藿、紫河车、当归、白芍、香附、红花、川芎、桃仁)对雄激素致无排卵大鼠模型血浆及卵巢血管舒张因子一氧化氮(NO)、环磷酸鸟苷(cGMP)、神经型一氧化氮合酶(nNOS)、内皮型一氧化氮合酶(eNOS)、血管收缩因子内皮素-1(ET-1)、血管紧张素Ⅱ(ATⅡ)的影响,探讨其治疗无排卵性疾病的作用及机理。方法:建立雄激素致无排卵大鼠模型。40只大鼠分为正常组、模型组和中药治疗组。采用放射免疫法测定大鼠血浆NO、cGMP、ET-1、ATⅡ以及血清雌二醇(E2)、孕酮(P)、睾酮(T)含量;采用免疫组化法检测大鼠卵巢nNOS、eNOS、ET-1表达;采用HE染色观察大鼠卵巢组织形态学变化。结果:与模型组比较,中药治疗组大鼠血浆NO、cGMP含量升高(P<0.01),血浆ET-1、ATⅡ含量降低(P<0.01),血清E2、T、E2/P降低(P<0.01);卵巢颗粒细胞、髓质nNOS、eNOS表达增加(P<0.01),ET-1表达降低(P<0.01);卵巢结构有一定改善,卵泡发育并有黄体形成。结论:补肾调经方具有一定的调节无排卵大鼠模型血浆及卵巢血管舒-缩因子、改善无排卵大鼠模型卵巢血液供应、抑制颗粒细胞凋亡和卵泡闭锁的作用,揭示其可能促进卵泡发育、成熟、排卵的作用机制。AIM:To observe the effect of BushenTiaojing Recipe (Radix rehmanniae preparata,Fructus lycii, Fructus rubi,Herba epimedii,Hominis placenta,Radix angelicae sinensis,Radix Paeoniae alba,Rhizoma cyperi, Flos carthami,Rhizoma chuanxiong and Semen persicae) of nitric oxide (NO),cyclic guanosine monophosphate (cGMP),neuronal nitric oxide synthase(nNOS),endothelial nitric oxide synthase(eNOS) of the vasorelaxation factors,and endothelin-1(ET-1) and angiotensin Ⅱ(ATⅡ) of vasoconstriction factors in androgen-induced anovular rats on plasma and ovarian,exploring the action and mechanism for treating anovular diseases. METHODS:The anovular model rats induced by injecting the female rats testosterone. Forty rats were divided into normal group,model group and herbs-treated (BushenTianjin Recipe) group,the radioimmunoassay was used to detect the contents of plasma NO,cGMP,ET-1,ATⅡ and serum estradiol (E2),progesterone(P)and testosterone(T),immunohistochemical method was applied to measuring the expression of ovarian nNOS,eNOS and ET-1,H-E staining was used to observe the changes in ovarian histomorphology. RESULTS:Compared with the rats in model group, the plasma NO,cGMP increased(P 0. 01),and plasma ET-1,ATⅡ decreased(P 0. 01),the serum E2,T,E2 / P decreased in the herbs-treated rats(P 0. 01); the expression of nNOS、eNOS increased(P 0. 01)and ET-1 decreased in granular cell and ovarian medulla in the herbs-treated rats; the ovarian structure was improved to some extent,there were follicular development and formation of corpus luteum in the herbs-treated rats. CONCLU-SION: BushenTiaojing Recipe may regulate the plasma and ovarian vasomotion factors,improve the ovarian blood supply inhibit apoptosis of granular cell and follicular atresia in androgen-induced anovular rats. The study reveals BushenTiaojing Recipe’s possible action mechanism to promot the follicular development,mature and ovulation.
