巴戟天药物血清对成骨细胞生物学特性的影响

摘　　要：目的:通过观察巴戟天药物血清对体外培养成骨细胞(OB)增殖及其活性的影响,探讨巴戟天壮骨作用的实质。方法:取新生SD大鼠头盖骨提取成骨细胞,给巴戟天药物血清,测定巴戟天对成骨细胞(OB)增殖、分泌碱性磷酸酶(ALP)、骨钙素(OC)及转化生长因子(TGF-β1)的影响。结果:巴戟天醇提物及巴戟天正丁醇部位能显著刺激OB增殖;巴戟天中正丁醇部位低剂量组可促进分化中的OB分泌ALP和OC,并可促进OB表达TGF-β1。结论:巴戟天中正丁醇部位低剂量组能够促进OB分泌碱性磷酸酶、表达TGF-β1mRNA,从而促进骨的生长。Objective：To explore the essence of Radix Morindae Officinalis in strengthening bones through observing the effect of medicated serum of Radix Morindae Officinalis on cultured osteoblast in vitro. Methods：The medicated serum of Radix Morindae Officinalis was added to the osteoblasts which were separated from the cranium of 24-hours newborn SD rat,in order to determining the effect of TGF-β1 and the proliferation and secreting of alkaline phosphatase and osteocalcin of the osteoblasts. Results：The proliferation of the osteoblasts were signifi cantly stimulated by the alcohol and the n-butanol extract from the Radix Morindae Officinalis; and the n-butanol extract from the Radix Morindae Officinalis could promote differentiating OB excreting the ALP and OC;in addition,the ability of the osteoblasts expressing TGF-β1 was promoted by the n-butanol extract from the Radix Morindae Officinalis. Conclusion：The n-butanol extract from the Radix Morindae Officinalis can promote the ability of the osteoblast in expressing TGF-β1 mRNA and excreting the ALP,and further promote the growth of bone.
