摘 要:目的:建立免疫性血小板减少性紫癜(ITP)小鼠动物模型,观察益髓颗粒对模型小鼠外周血细胞因子影响,探讨益髓颗粒的免疫调节机制。方法:40只Balb/c小鼠随机分为正常组、模型组、泼尼松治疗组和益髓颗粒治疗组,除正常组外,其余各组隔日1次腹腔注射豚鼠抗小鼠血小板血清(GP-APS)建立ITP小鼠模型,于造模第8d开始,各组均按0.2mL/(10g·d)体积灌胃。其中正常组、模型组灌服生理盐水;泼尼松按0.0113mg/(10g·d)灌胃;益髓颗粒按0.3g/(10g·d)灌胃。连续7d后眼球取血分离血清,酶联免疫吸附法(ELISA)检测干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)。结果:与正常组相比,模型组小鼠IFN-γ水平明显升高,IL-4水平明显降低,IFN-γ/IL-4比值明显异常;与模型组相比,泼尼松治疗、益髓颗粒治疗组的IFN-γ、IL-4均有所恢复,IFN-γ/IL-4比值接近正常。结论:益髓颗粒可通过影响Th1/Th2细胞及其因子水平而对ITP免疫功能起调节作用。Objective:To observe the effects of Yisui Granule on peripheral blood cytokines in immune thrombocytopenic purpura (ITP) mouse model and discuss it's immune regulatory mechanism. Methods:40 Balb/c mice were randomly divided into normal group,model group,prednisone acetate group and Yisui Granule group. ITP mouse model were made by injected intraperitoneally guinea pig-antimouse platelet serum(GP-APS) every other day. On the 8th day establishing the model,each group were inrtagastrically at a dose of 0.2mL/(10g·d) volume. Normal group and model group were fed with normal saline,prednisone acetate and Yisui Granule were inrtagastrically administered for seven days at a dose of 0.0113mg/(10g·d),0.3g/(10g·d) respectively. The levels of IFN-γ and IL-4 in peripheral blood serum were detected by ELISA. Results:The level of IFN-γ was significantly higher in model group than that in normal group,and the levels IL-4 decreased in model group,the ratio of IFN-γ/IL-4 increased. Compared with model group,the levels of IFN-γ and the ratio of IFN-γ/IL-4 decreased,the levels of IL-4 increased in prednisone acetate group and Yisui Granule group. Conclusion:Yisui Granule can regulate the immune function of ITP by influencing the level of Th1/Th2 cells and their cytokines.
