强心益气汤对多柔比星所致急性心衰大鼠心功能的保护作用

摘　　要：目的:观察强心益气汤对多柔比星致急性心衰大鼠心功能的影响及机制。方法:一次性腹腔注射多柔比星10mg/kg,复制急性心衰大鼠模型。强心益气汤灌胃6d后,观察心室内压变化、血清中超氧化物歧化酶(super oxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)含量的影响以及观察心肌病理形态学的改变。结果:强心益气汤各组均能不同程度改善血流动力学并能明显升高血清SOD、降低MDA含量;HE染色结果显示,强心益气汤各组心肌细胞变性坏死明显减轻。结论:强心益气汤能够明显改善多柔比星心肌毒性导致的急性心衰,该作用可能与调节大鼠体内自由基代谢及增强心脏泵血功能有关。Objective：To observe the protective effects and mechanisms of Qiangxin Yiqi Decoction（QXYQD） on heart function of rats with doxorubicin-induced acute heart failure.Methods：SD rats were randomly divided into 4 groups：normal control （NC） group,model control （MC） group,low dosage of QXYQD （L-QXYQD） group,high dosage of QXYQD （H-QXYQD） group.QXYQD was repeatedly administered at dose of 24,72g/kg,respectively,1 time a day for 6d.On the 7th day,the model control and QXYQD groups were administered with doxorubicin （20mL/kg） through the intraperitoneal injection once to cause heart failure.MS4000U-1C biological signal collecting system was used to record and analyze the hemodynamics parameters of the rats,examine the contents of SOD and MDA in serum.The changes of cardiac pathological histology were observed under microscope.Results：Compared with NC group,the LVSP,lv ＋ dp/dtmax,the contents of SOD in serum of the MC group significantly lowered （P0.01）,but in the L-QXYQD group and H-QXYQD group those index were markedly higher than those of the MC group （P0.05）.In the MC group,marked myofibrilla cellular edema,degeneration and necrosis of myocardial tissue were found under microscope.The myocardial necrosis was markedly alleviated in L-QXYQD group and H-QXYQD group,compared with that of the MC group.Conclusion：QXYQD can improve the changes of cardiac pathological histology of rats with doxorubicin-induced acute heart failure,which may partly depend on regulating oxyradical in body and preserving cardiac myofibrilla.
