摘 要:接种于含NaCl 培养基的无花果愈伤组织细胞生长极显著受抑,Na+ 含量增加,K/Na 比值下降,游离脯氨酸积累。培养基中添加一定量CaCl2 不仅在一定程度上缓解盐分对生长的抑制作用,增加K+/Na + 比,而且明显促进游离脯氨酸积累。如果在添加钙的同时再添加细胞钙调素活性抑制剂盐酸氯丙嗪(CPZ) 或盐酸三氟拉嗪(TFP) ,均使钙促进的脯氨酸积累受到明显抑制,表明盐胁迫诱导的脯氨酸积累可能涉及细胞CaCaM系统。The growth of fig (Ficus carica L.) calli cultured on MS media was sharply inhibited by NaCl(Fig.1) with dramatical increase in sodium contents and decrease in K/Na ratio, in addition to proline accumulation. Addition of calcium to media alleviated the inhibition of growth by salt stress, lowered sodium contents(Fig.2) and stimulated free proline accumulation(Fig.3). Addition of the calcium chelating agent EGTA, calcium channel blocker CoCl 2 and CaM inhibitors CPZ and TFP to culture media containing higher calcium concentrations under NaCl stress depressed to different degrees the salt induced proline accumulation promoted by calcium(Fig.5). The results showed that CaM activation might be necessary in calcium promotion of the accumulation of proline in fig calli.
