水稻光敏素基因(phyA)和-1,5-二磷酸核酮糖羧化酶/加氧酶小亚基基因(rbcS)的染色体定位

摘  要:以生物素标记的水稻单拷贝光敏素基因(phyA) 和1 ,5二磷酸核酮糖羧化酶/ 加氧酶小亚基基因(rbcS) 的基因组克隆为探针,其大小分别为6 .6 和1 .1 kb ,通过原位杂交技术将它们分别定位到水稻染色体上。phyA 和rbcS基因的检出率分别为29 .79 % 和21 .56 % 。phyA在第3 染色体上有3 个座位:长臂近着丝粒、短臂末端、长臂中部。rbcS分别定位于第7 染色体长臂近着丝粒(8 .62 % ) 、第5 染色体长臂末端、第6 染色体长臂距着丝粒近2/3 处。此外,对信号转导相关基因定位的意义,水稻染色体的准确识别、功能基因在染色体上的分布及位置意义等也进行了讨论。Phytochrome is the most important photoreceptor protein, ribulose 1,5 bisphosphate carboxylase (Rubisco) small subunit (SSU) genes ( rbcS ) are regulated developmentally by light. The genes coding for phytochrome and Rubisco SSU are functionally related. In order to understand the relationship between their structure and function in rice genome and the differences between the genetic map and the physical map, rice phyA gene clone, pPHY1 and rbcS gene clone, pRR1 were physically mapped by biotin labelled in situ hybridization. The results showed that the hybridization detection ratio was about 29.79% for phyA and 21.56% for rbcS (Table 1). The gene phyA was located near the centromere of the long arm, at the end of the short arm and at the middle of the long arm on the chromosome 3 (Fig.1, Plate Ⅰ A~D). The gene rbcS was mapped near the centromere on the long arm of chromosome 7, at the end of the long arm of chromosome 5, and at the 2/3 distance of the long arm from the centromere in chromosome 6. It is suggested that the fragments for phyA gene be dispersed in chromosome 3 (Fig.1, PlateⅠ F~I). Our results of physical mapping of multiple positions of rbcS in chromosomes are consistent with previous results. The significance of localizing genes related to signal transduction, the identification of rice chromosomes, and the understanding of the meaning of position and distribution of functional genes along the chromosomes were also discussed.
