何首乌饮对衰老雄性大鼠睾丸生殖功能的影响

摘　　要：目的:探讨何首乌饮对衰老雄性大鼠生殖功能的影响。方法:32只雄性SD大鼠随机分为正常组、衰老模型组、何首乌饮组和何首乌丸组,每组8只。采用D-半乳糖连续腹腔注射建立亚急性衰老大鼠模型,何首乌饮组和何首乌丸组大鼠分别给予何首乌饮(38.4g/kg/d)和何首乌丸(8.24g/kg/d)灌胃60d。光镜下观察大鼠睾丸附睾头精子数量、存活率和活动度,放射免疫法检测血清睾酮浓度,原位杂交法观察睾丸IGF-1 mRNA的表达。结果:何首乌饮可显著提高衰老大鼠精子数量、存活率和活动度,血清睾酮浓度,睾丸IGF-1 mRNA的表达;何首乌饮组大鼠各指标与何首乌丸组、模型组比较差异显著(P<0.05)。结论:何首乌饮可改善衰老雄性大鼠的生殖功能,延缓性腺衰老。Objective：To investigate the effects of Heshouwuyin on reproductive function of aging male rats.Methods：32 male SD rats were randomly divided into 4 groups（n=8）：normal control group,aging model group,Heshouwuyin group and Heshouwuwan group.The subacute aging rat model was made by injecting Dgal into the abdominal cavity continually.After successfully establishing the aging rat model,the rats in Heshouwuyin group and Heshouwuwan group were respectively lavaged with Heshouwuyin（38.4g/kg/d） and Heshouwuwan（8.24g/kg/d） for 60 days.The caput epididymidis sperm count,vitality and survival rate of sperms were observed by microscopy;Radioimmunoassay was used to detect serum testosterone concentration,in situ hybridization to observe the expression of IGF-1 mRNA in testis.Results：Heshouwuyin could obviously increase sperm count,vitality and survival rate of sperm,serum testosterone concentration and IGF-1mRNA in testis of model rats.All indexes of rats in Heshouwuyin group had obvious differences with those of rats in Heshouwuwan group and model group（P〈0.05）.Conclusions：Heshouwuyin can improve the reproductive function of aging male rats,and delay gonad aging.
