摘 要:合成了带正电荷密度不同的聚离子亚基化合物(ionenepolymers),探讨了聚合物结构对外源基因转染真核细胞的影响规律。从实验结果看,聚离子亚基化合物结构影响外源基因转染效率,但其影响程度与靶细胞的种类相关;对HeLa细胞、NIH3T3细胞,聚离子亚基化合物的正电荷密度加大,其促基因转移功能加强;而对PA317包装细胞,结果相反。实验结果还表明,阳离子聚合物浓度过高或过低也会降低外源基因的转染效率。Using synthetic and different positive charge density ionene polymers and the effect law of their structures on transient expression of exogenous genes in eukaryocytes were determined.The results showed the structures of the ionene polymers affected the efficiency of exogenous transient transformation, the levels of effect were associated with the kinds of target cells. In HeLa and NIH3T3 cells, with the increasing of positive charge density of ionene polymers, the function of their promoting gene transfer enhanced, but in PA317 package cells, the results were opposite. The results also indicated that the extra high or low concentrations of ionene polymers reduced the efficiency of exogenous gene transfer.
