摘 要:目的:研究在灌服蛤蚧乙醇提取液去势大鼠胫骨中TGF-β1的表达,探讨蛤蚧乙醇提取液防治绝经后骨质疏松症的机理。方法:采用12月龄SD雌性大鼠60只,随机分为对照组、去势组和实验组。去势组和实验组均切除双侧卵巢,实验组给予蛤蚧乙醇提取液灌胃处理;对照组给予假手术处理。3组大鼠分别于术后4、12周,随机各取10只,测定骨密度(BMD)后处死,取右侧胫骨,脱钙后包埋切片,HE染色,计数破骨样细胞;常规SABC法进行免疫组化染色,图像分析灰度值。结果:实验组的全身BMD明显高于去势组,但仍然低于对照组;实验组的破骨样细胞计数与对照组相似,与去势组比较差异有统计学意义(P<0.05)。TGF-β1在实验组及对照组的成骨细胞中呈阳性染色,部分骨基质中也有阳性表达,去势组呈阴性表达。结论:蛤蚧乙醇提取液可能诱导骨微环境中TGF-β1表达增加,进而抑制破骨样细胞的生成,有效地预防绝经后骨质疏松的发生。Objective: To study the expression et al. Medical of TGF β1 in the tibia of ovariectomized rats with gastric perfusion of Gekko gecko ethanol-extracts, and discuss the mechanisms of Gekko gecko ethanol-extracts to prevent postmenopausal osteoporosis. Methods:Sixty female SD rats at the age of 12 months were randomly divided into control group, tendency group and experimental group. Ovaries of tendency group and experimental group were removed. The control group was sham-operated. The experimental group was given Gekko gecko ethanol-extracts at the same time. Four and 12 weeks after operation, 10 rats of each group were killed, their right tibias were taken, decalcified,made into embedded sections, some were stained with HE and counted osteoclast-like cells. Some were stained with immunohistochemical method, and analysised the gray value of TGF-β1. Results:The number of osteoclast- like cells in experimental group was similar to that in contol group, but was significant difference from that in tendency group. There were TGF-β1 positive osteoblasts in control group and experimental group, some TGF-β1 positive cells were also found in bone matrix, but TGF-β1 showed negative expression in tendency group. Conclusion.Gekko gecko ethanol-extracts could induce the expression of TGF-β1 and inhibit the generation of osteoclast-like cells in bone microenvironment, so prevent the occurrence of postmenopausal osteoporosis effectively.
