摘 要:目的:观察不同剂量^(60)Co-γ射线照射后大鼠海马突触超微结构的改变,探讨^(60)Co-γ射线照射后导致学习记忆能力下降的机理。方法:将成年SD大鼠随机分为正常对照组、10Gy照射组、30Gy照射组,采用Morris水迷宫测试大鼠学习记忆活动,用透射电镜观察海马CA3区突触超微结构。结果:随着照射剂量的增加,大鼠第1次穿越目标区的时间逐渐延长,海马CA3区突触数量和突触后致密物厚度也逐渐减少;与正常对照组相比,30Gy照射组突触间隙显著增宽,穿越目标区的次数显著减少,但与10Gy照射组比较无明显差异(P>0.05),两照射组间也无差异(P>0.05)。结论:^(60)Co-γ射线照射可引起大鼠海马突触丧失和结构改变,导致学习记忆能力下降。Objective: To observe the ultrastructural changes in Hippocampus' Synapses of the rats which were irradiated with different doses of ^60Co-γ ray irradiation, to explore the mechanism leading to decreased learning and memory ability after radiation. Methods:Adult Sprague-Dawley rats were divided into normal control group, 10 Gy-irradiated group and 30 Gy-irradiated group. The state of learning and memory activities was tested by Morris water maze, and the ultrastructure of hippocampus' synapses in CA3 region of the rats were observed under electron microscope. Results:With the irradiation increasing, the first time of the rats swimming through the plateau was increased, while the number of synapse and the thickness of postsynaptic density were decreased. The widths of synaptic cleft and the times through the plateau were more significantly increased in 30 Gy-irradiated group than those in control group. But there was no difference in the widths of synaptic and the times swimming through the plateau between 10 Gy group and normal control group( P 〉0.05), and it was the same with the 30 Gy group and 10 Gy group( P 〉0.05). Conclusion:^60Co-γ ray irradiation injures hippocampus and causes the decreasing of the number and the ultrastruetural abnormalities of the synapse and decreases learning and memory ability in the rats.
