缺氧相关因子HIF-1α、AQP-1表达与宫颈鳞状细胞癌血管生成关系的研究

摘  要:目的:探讨HIF-1α、AQP-1表达对宫颈鳞状细胞癌血管生成的作用。方法:应用免疫组织化学方法检测54例宫颈鳞状细胞癌中HIF-1α、AQP-1的表达,用CD105标记肿瘤微血管密度。结果:HIF-1α在宫颈鳞状细胞癌的阳性表达率为50%,表达水平与分化程度、浸润深度明显相关(r=-0.674,P<0.05和r=0.897,P<0.01),与临床分期、肿瘤体积、淋巴结转移无明显相关(P>0.05)。AQP-1在宫颈鳞状细胞癌的表达率为87.1%,表达水平与肿瘤体积、临床分期明显相关(r=0.989,P<0.05和r=0.807,P<0.01),与分化程度、淋巴结转移、浸润深度无明显相关(P>0.05)。微血管密度与肿瘤体积、浸润深度明显相关(r=0.609,P<0.05和r=0.845,P<0.01);与临床分期、分化程度、淋巴结转移无明显相关(P>0.05)。微血管密度与HIF-1α表达明显相关(r=0.701,P<0.05),而与AQP-1的表达无关(P>0.05)。AQP-1的表达水平与HIF-1α的表达水平明显相关(r=0.648,P<0.05)。结论:HIF-1α、AQP-1均参与宫颈鳞状细胞癌缺氧诱导新生血管过程,并且与微血管密度密切相关,其中HIF-1α发挥重要作用。Objective:To explore the effection of HIF-1α,AQP-1 on angiogenesis in cervical squamous cell carcinoma.Methods:The expressions of proteins HIF-1α,AQP-1 were detected by immmunohistochemistry method in 54 cases of cervical squamous cell carcinoma.The micro vessel density(MVD) were detected by CD105.Results:The positive expression ratio of HIF-1α in cervical squamous cell carcinoma was 50%.Expression of HIF-1α related to tumor differentiation,invasive depth(r=-0.674,P〈0.05;r=0.897,P〈0.001),but not to tumor volume,clinical stage,lymphonode metastasis(P〉0.05).The positive expression ratio of AQP-1 was 87.1%.The expression of AQP-1 had relationship with tumor volume,clinical stage(r=0.989,P〈0.05;r=0.807,P〈0.001),but not with the tumor differentiation,invasive depth and lymphonode metastasis(P〉0.05).There was relationship between the expression of HIF-1α and tumor microvessel density(r=0.701,P〈0.05).There was no relationship between the expression of AQP-1 and tumor microvessel density(P〈0.05).The tumor microvessel density had relationship with tumor volume,invasive depth(r=0.609,P〈0.05;r=0.845,P〈0.001),but had no relationship with tumor differentiation,clinical stage,lymphonode metastasis(P〈0.05).There was relationship between the expression of HIF-1α and AQP-1(r=0.648,P〈0.05).Conclution:HIF-1α,AQP-1 both take part in the procession of angiogenesis induced by hyposia in cervical squamous cell carcinoma and have relationship with tumor microvessel density;HIF-1α plays key role in the whole procession.
