摘 要:目的:探讨骨髓间充质干细胞(BMSCs)体外向子宫内膜上皮细胞方向分化的可行性。方法:原代培养小鼠BMSCs和子宫内膜间质细胞(ESCs)并鉴定,将BMSCs与ESCs共培养,并补充生长因子(TGF-β10ng/ml,EGF10ng/ml,PDGF-BB10ng/ml)和雌激素(17-β-雌二醇1×10-7mol/L),应用细胞免疫荧光染色法检测分化的BMSCs是否表达上皮细胞特异标记角蛋白。结果:小鼠BMSCs增殖潜能强,呈克隆性生长,其表面标记Sca-1,CD117,CD29,CD34表达率分别为5.36%,2.41%,2.63%,0.68%。小鼠ESCs特异标记波形蛋白表达阳性,角蛋白阴性。小鼠BMSCs与ESCs共培养5天后大部分细胞形态由成纤维样向上皮样细胞变化,细胞免疫荧光染色方法鉴定诱导分化后的BMSCs表达角蛋白。结论:小鼠BMSCs在ESC分泌和外源性因素诱导下可向子宫内膜上皮细胞方向分化。生长因子和雌激素在此过程中起重要作用。Objective:To explore the plasticity of differentiation of bone mesenchymal stem cells(BMSCs) into endometrial epithelial cells in vitro.Methods:Mice's BMSCs and endometrial stromal cells(ESCs) were isolated and identified in certain ways.The BMSCs were induced by co-cultured with ESCs,added 10ng/ml TGF-β,10ng/ml EGF,10ng/ml PDGF-BB,1×10-7mol/L β-estradiol.The expression of cytokeratin which was sign of endometrial epithelial cells was tested by immunofluorescence in differentiated BMSCs.Results:BMSCs in vitro showed great potential of proliferation.The superficial markers of BMSCs:Sca-1,CD117,CD29,CD34 were 5.36%,2.41%,2.63%,0.68% respectively,indicating the cultured cells were BMSCs.After co-cultured with ESCs for five days,most differentiated BMSCs looked like epithelial cells.The differentiated BMSCs expressed cytokeratin,indicating that BMSCs had been differentiated into endometrial epithelial cells to some extent.Conclusion:BMSCs has the potential to differentiate into endometrial epithelial cells in vitro.The exogenous growth factors and estradiol play an important role.
