摘  要:目的观察电针对脑缺血大鼠皮质缺血半影区Bcl-2蛋白质表达的影响,为探讨电针治疗脑梗塞的机理提供实验依据。方法线栓雄性SD大鼠大脑中动脉建立脑梗塞动物模型,随机分为电针组和非电针组。用免疫组化的方法显示大脑皮质缺血半影区Bcl-2阳性细胞并观察细胞形态和分布,计数阳性细胞数量;用Western-blot方法定量皮质缺血半影区Bcl-2蛋白表达的变化。结果①对照组和假手术组大鼠脑皮质未显示出Bcl-2免疫阳性细胞。非电针组的大鼠皮质缺血半影区在缺血6h即在皮质中间带见圆形,偶见椭圆形和梭形Bcl-2免疫阳性细胞分布。阳性细胞的数量在缺血1d最多,3d呈减少的趋势。电针组的阳性细胞形态与分布与非电针组比未见明显差异,在数量上亦随电针的时间增加而增多。②Western-blot方法未在对照组和假手术组大鼠额叶皮质检出Bcl-2蛋白质。电针组大鼠皮质缺血半影区的Bcl-2的表达比非电针组的呈上调的趋势。结论电针刺激内关和足三里穴位可以上调脑梗塞大鼠皮质缺血半影区内Bcl-2蛋白质的表达,参与脑缺血性细胞凋亡的病理过程。Objective To provide experimental evidence for studying mechanism of electroacupuncture treating cerebral infarction. Methods Rat model of cerebral ischemia was established with the method of thread middle cerebral artery occlusion. And then the model rats were randomly divided into electroaeupuncture group and non-electroacupuncture group. At the same time the control group and sham group were matched in which the rats were only exposed middle cerebral artery but not occlusion. Immunochemistry was used to show Bcl-2 positive cells in the cerebral cortex, and the morphology and number of the posi- tive cells were investigated under optical microscopy as well as level of bcl-2 expression with Western-blot method. Results Bcl-2 positive cells were not found at cerebral cortex in the control and sham group by immunohistochemistry. In nonelec- troacupuncture group bcl-2 positive cells were showed at penambra region after ischemia 6 h and bcl-2 expression was peak at 1 day after ischemia and was less at three day than at 1 day, almost disappeared at 7 day. The morphology of positive cells in nonelectroacupuncture group was mainly round and ellipse or fusiform in shape, and the distribution of the positive cells was mainly at the middle region of cortex. In electroaeupuncture group, the morphology and distribution of positive cells were not sbouwed difference from nonelectroaeupuneture group, but the number of the positive cells was increased in electroacupunture group than non-electroacupuncture group. The level of bcl-2 expression was higher in eleetroacupuncture group than non-elec- troacupuncture group. Conclusions Electoacupuncture on Neiguan and Zusanli acupoint can up-regulate bcl-2 protein expression at penambra region of cerebral cortex in middle cerebral artery occluded rats.
