摘  要:目的:研究人精子囊性纤维化跨膜转导调节因子(cystic fibrosis transmembrane conduc-tanceregulator,CFTR)表达率与精子受精能力间的关系。方法:运用间接免疫荧光法及蛋白印迹分析法研究CFTR在人精子上的表达及其在不同类型(生育组、不明原因性不育组及不育组)精子上的表达率情况。结果:通过间接免疫荧光法发现CFTR表达在人精子赤道板上,蛋白印迹分析进一步证实人精子中存在CFTR,分子量为170kD;采用间接免疫荧光技术,CFTR在生育组、不明原因性不育组及不育组(包括畸形精子症、弱精症、少精症)上的表达率有显著差异:CFTR在生育组精子上的表达率(88.13%±3.79%)远高于不明原因性不育组(52.93%±15.36%)及不育组(35.89%±20.41%)(P<0.01)。结论:CFTR表达率下降与人精子受精能力降低有关。Objective: To investigate the correlation between percentage of human sperm expressing CFTR and sperm fertilizing capacity.Methods: The expression of CFTR on spermatozoa was conducted by indirect immunofluorescence staining and Western Blot Analysis.And the percentage of spermatozoa expressing CFTR from fertile,unexplained infertile and infertile men (mainly teratospermic,asthenospermic and oligospermic) was examined with indirect immunofluorescence staining.Results: CFTR protein was localized to the equatorial segment of sperm head and molecular weight was approximately 170 kD.And the percentage of spermatozoa expressing CFTR in fertile men was significantly different from that in the unexplained infertile men and infertile men categories,the fertile men spermatozoa greatly higher (88.13% ±3.79%)than that the unexplained infertile men (52.93% ±15.36%) and infertile men (35.89% ± 20.41%) (P 0.01).Conclusion: The defective expression percentage of CFTR in human sperm is related to a reduction of sperm fertilizing capacity.
