摘 要:目的探讨脑益康对阿尔茨海默病(AD)模型大鼠学习、记忆能力和脑内炎性反应的影响。方法采用β-淀粉样蛋白1~40(Aβ1~40)右侧海马注射制备AD模型大鼠,脑益康灌胃给药28d后,采用Y-型迷宫刺激器检测大鼠学习、记忆能力,免疫组织化学方法检测大鼠海马肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)的表达情况,酶联免疫吸附法(ELISA)检测皮层白细胞介素-1β(IL-1β)、转化生长因子-β1(TGF-β1)的蛋白水平。结果脑益康能改善模型大鼠学习记忆能力,显著降低AD大鼠脑组织的IL-1β、IL-6、TNF-α的含量(P<0.01),增加TGF-β1的表达(P<0.01)。结论脑益康对AD模型大鼠学习记忆能力有改善作用,通过降低AD大鼠脑组织的IL-1β、IL-6、TNF-α的含量,增加TGF-β1的表达,有效抑制海马神经元炎性反应。Objective To investigate effects of Naoyikang on improving learning and memory ability and inflammation reaction of Alzheimers′ disease (AD) model rats. Methods AD model rats were established by the injection of Aβ1~40 into the right hippocampus. After intragastrically administrated with different solution for 28 days,learning and memory abilities of rats were tested by Y-maze. The levels of TNF-α and IL-6 in AD rats′ hippocampus were detected by immunohistochemistry. The levels of IL-1β、TGF-β1 in cortex were measured by ELISA. Results Naoyikang could improve the learning and memory abilities of AD rats,descend significantly the concentration of IL-1β、IL-6 and TNF-α (P0.01),and increase the expression of TGF-β1(P0.01).Conclusions Naoyikang can improve the learning and memory abilities of AD rats,and effectively control the inflammation reaction by decreasing the content of IL-1β,IL-6 and TNF-α and increasing the expression of TGF-β1.
