摘  要:目的对微生物管碟法与比浊法测定麦白霉素效价与组分的关系加以探讨研究,说明比浊法测定效价较管碟法测定效价的高灵敏度与高选择性。方法比浊法以金黄色葡萄球菌为实验菌,加菌量约0.3%(V/V),(37±0.5)℃培养3.5-4.5h后测定吸光值。管碟法按照中国药典2005年版二部。HPLC法:色谱柱为phenomenex Gemini C18(4.6mm×250m);流动相为0.2mol/L甲酸铵溶液(pH7.6)-乙腈(60﹕40);流速为1.0mL/min;检测波长为232nm;柱温为30℃;进样量为10μL。结果比浊法更好地反映了麦白霉素组分的变化,管碟法对组分的变化基本无反应。结论比浊法较管碟法对组分变化的反应灵敏度高、选择性好,更好地反应了样品的实际质量。Objective To Study on relation between the content of microbial assays (the cylinder-plate assay and the turbidimetric assay) and the component of meleumycin, thus to reveal the better selection of turbidimetric assay than the cylinder-plate assay. Methods The turbidimetric assay :The test organism was Staphylococcus aureus. Adjusted bacterium suspension of Staphylococcus aureus inoculated with a about 0.3% (V/F) ratio, at (37±0.5)℃ for 3.5-4.5 hours. The cylinder-plate assay: Chinese .Pharmacopeia 2005 Vol 2. HPLC method: The analytical column was phenomenex Gemini C18 (4.6mm×250mm, 5μm). The column temperature was 30℃. The mobile phase was consisted of the 0.2mol/L ammonium formate solution (pH7.6)-acetonitrile (60 : 40). The flow rate was 1.0mL/min. The detection wavelength was 232nm and the inject volume was 10μL. Results The result of the turbidimetric assay reflect the change of the meleumycin component bettly, at the meantime the cylinder-plate assay could not do this. Conclusion The turbidimetric assay was more specific than the cylinder -plate assay to reflect the actual change of the meleumycin component.
