摘 要:目的:建立快速筛查呼吸道标本的实时荧光定量PCR法,考察其在呼吸道病原体快速筛查中的应用。方法:采用国家标准细菌培养、病毒培养、实时聚合酶链反应(PCR)技术对2007年-2009年12月份丽水市10起常见呼吸道疫情的232份样本进行检测,比较方法的灵敏度、特异性及准确性。结果:166份样本的细菌培养与实时荧光定量PCR检测结果符合率为62.50%,其中6份普通细菌培养阴性,PCR阳性。66份样本的病毒培养与实时荧光定量PCR检测结果符合率为88.89%。结论:实时荧光定量PCR不但检测速度快,操作方便,而且具有准确、灵敏度高、特异性好的优点。Objective:To establish a method of fluorescent quantitative PCR and examine its utility for rapid pathogens′ screening in respiratory infectious disease.Methods:232 specimens of 10 respiratory infectious disease outbreak events occurred in Lishui during the year of 2007-2009 were detected by culture method of national standards and compared with the method of fluorescent quantitative PCR.The results of different methods were compared and the sensitivity,specificity and accuracy of fluorescent quantitative PCR method was obtained.Results:The detective result of fluorescent quantitative PCR was 62.50% in accordance to that of common bacterial culture during 166 samples.6 specimens were detected negative by bacterial culture were positive by fluorescent quantitative PCR method.The detective result of fluorescent quantitative PCR was 88.89% accordance to that of viral culture during 166 samples.Conclusion:Real-time fluorescent quantitative PCR is an ideal method for rapid pathogens′ screening in respiratory infectious disease with high accuracy and sensitivity.
