摘 要:目的:观察通心络对自发性糖尿病(GK)大鼠心肌Ⅰ型和Ⅲ型胶原阳性表达及含量、细胞转化生长因子(TGF-β1)含量的影响。方法:4月龄GK大鼠30只,随机分为GK对照组、通心络大、小剂量组,另取4月龄Wistar大鼠10只作正常对照,分别灌胃12周。灌胃结束后,制备心肌切片,免疫组化染色,计算胶原I、胶原Ⅲ阳性面密度比值以代表表达程度;取心肌匀浆,酶联免疫方法测定心肌Ⅰ、Ⅲ型胶原及TGF-β1含量。结果:与同月龄正常大鼠相比较,GK大鼠血糖明显增高(P<0.01),通心络药物组有降低趋势,但无差异(P>0.05)。与正常大鼠比较,GK大鼠心肌胶原Ⅰ阳性面密度比值明显增高(P<0.01),胶原Ⅲ阳性面密度比值明显降低(P<0.01),心肌匀浆胶原Ⅰ含量明显增高(P<0.01),胶原Ⅲ含量降低(P<0.05),心肌胶原Ⅰ/Ⅲ比值明显增高(P<0.01),TGF-β1含量降低(P<0.01);与7月龄GK大鼠比较,通心络组胶原Ⅰ阳性面密度明显降低(P<0.01),高、低剂量组之间有差异(P<0.05),胶原Ⅲ阳性面密度明显升高(P<0.01);胶原Ⅰ含量明显降低(P<0.01),胶原Ⅲ含量明显提高(P<0.01),胶原Ⅰ/Ⅲ比值降低(P<0.01),TGF-β1含量明显提高(P<0.01),高、低剂量组之间无差异(P>0.05)。结论:通心络可降低GK大鼠心肌胶原Ⅰ阳性表达,Ⅰ型胶原含量,升高胶原Ⅲ阳性表达,Ⅲ型胶原含量,降低胶原Ⅰ/Ⅲ比值,升高TGF-β1含量,具有改善心肌间质网络重构作用。Objective:To observe the effects of tongxinluo on myocardial Ⅰ,Ⅲ type collagen positive expression and TGF-β1 expression in GK rats.Methods:30 GK rats were divided into model,tongxinluo small and big dose groups at random,10 rats were taken as normal control,each group was given the corresponding medicine for 12 weeks.Rtas' myocardial Ⅰ,Ⅲ type collagen expression were determined by immunohistochemical method,and their contents and TGF-β1 content were counted by enzyme-linked immunoadsorbent assay(ELISA).Results:The blood glucose of GK rats was obviously higher than that of normal group,tongxinluo could lower blood glucose level,but there was not difference.The myocardial I type collagen positive areal density of GK rats was obviously higher than that of normal group,myocardial Ⅲ type collagen positive areal density was lower than that of normal;the myocardial homogenize I type collagen content of GK rats was increased and Ⅲ type collagen content was decreased,the ratio of Ⅰ/Ⅲ type collagen was raised,the myocardial homogenize TGF-β1 content was lowered.Tongxinluo could significantly lower myocardial I type collagen positive areal density,there was difference between the small and big dose groups;raise myocardial Ⅲ type collagen positive areal density;reduce the myocardial homogenize I type collagen content and elevate the Ⅲ type collagen content,decrease the ratio of Ⅰ/Ⅲ type collagen,increase TGF-β1 content,there was no difference between the two dose groups.Conclusion:Tongxinluo can improve myocardial collagen network remodeling,lowering myocardial I type collagen expression and content,raising Ⅲ type collagen expression and content,decreasing the ratio of Ⅰ/Ⅲ type collagen and increasing TGF-β1 content maybe the part of its mechanism.
