摘 要:目的探讨荣肝合剂对刀豆蛋白A(ConA)介导的急性免疫性肝损伤小鼠的保护作用及其机制。方法乙型肝炎病毒转基因小鼠60只,随机分为模型组、空白组、荣肝合剂组、茵陈蒿汤组、茵陈组、联苯双酯组,每组10只,采用ConA尾静脉注射法制作急性免疫性肝损伤模型。造模前14天,模型组、空白组给予生理盐水灌胃,其余组每日分别给予荣肝合剂、茵陈蒿汤、单味茵陈煎液、联苯双酯溶液灌胃。末次灌胃给药后1h,空白组给予磷酸盐缓冲液尾静脉注射,其余各组按照3μg/g体重尾静脉注射ConA造模。造模给药后8h处死动物取血或组织标本检测丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、总胆红素(TBiL)、乙型肝炎病毒载量(HBVDNA)、乙型肝炎表面抗原(HBsAg)、超氧化物歧化酶(SOD)、丙二醛(MDA)等指标。结果与模型组及其他给药组比较,荣肝合剂可降低小鼠ALT、AST、TBiL水平(P<0.05或P<0.01);与联苯双酯组、茵陈组、茵陈蒿汤组相比较,荣肝合剂可降低小鼠HBVDNA载量,降低肝组织MDA含量,提高肝组织的SOD水平,其余各组均无此作用。结论荣肝合剂的降酶作用优于单味中药茵陈、茵陈蒿汤及联苯双酯,其作用机制是通过抑制HBVDNA,减轻肝细胞膜脂质过氧化反应的损害,起到肝保护作用。Objective To investigate the protective effect and mechanism of Ronggan Mixture(Herbal Decoction for liver diseases)on the mice model with concanavalin A-induced acute immunological liver injury.Methods Totally 60 HBV transgenic mice were randomized into model group,control group,Ronggan Mixture group,Yinchenhao Decoction group,Yinchen group,and Bifendate group,with 10 in each.The acute immunological liver injury models were established by the tail vein injection of concanavalin A.Fourteen days before modeling,the model group and control group were given normal saline by gavage,while the other groups were treated with Ronggan Mixture,Yinchenhao Decoction(Oriental Wormwood Decoction),Yinchen(Virgate Wormwood Herb),and Bifendate respectively by gavage everyday.One hour after the last intervention,the control group was given phosphate buffer solution(PBS)by the tail vein injection,and the others were given concanavalin A by the tail vein injection to establish models.Eight hours after modeling,the mice were sacrificed,the serum and tissue samples were collected to detect:alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBiL),hepatitis B virus quantity(HBV DNA),hepatitis B surface antigen(HBsAg),superoxide dismutase(SOD),and malondialdehyde(MDA).Results Compared with the model group and the others,Ronggan Mixture could decrease the level of ALT,AST,and TBiL(P0.05 or P0.01).Compared with the Bifendate group,Yinchen group,and Yinchenhao Decoction group,Ronggan Mixture could decrease the HBV DNA and the MDA of liver tissues,and increase SOD level of liver tissues.Conclusion Ronggan Mixture is of advantage in decreasing the enzymes as compared with Yinchen,Yinchenhao Decoction and Bifendate.The liver protective effect is achieved through inhibiting HBV DNA to reduce the injury of the lipid peroxidation of hepatocyte membrane.
